PEG/NaCl

Polyethylene Glycol (PEG)/NaCl (500 mL)

PEG/NaCl is used as a precipitation agent in the purification of phage display libraries, individual phage clones, and other phage solutions. Phage are often amplified and purified in between rounds of selection in biopanning protocols.

Reagents


100 g Polyethylene glycol (PEG) 8000

116.9 g NaCl

Protocol


- Mix 100 g PEG, 116.9 g NaCl, and 475 mL water.


- Stir until the solutes dissolve, which may take about 1 h.

-It may be necessary to briefly heat the solution to 65°C to dissolve the solutes.

-Optional: The solution can be autoclaved. As it cools, mix the solution by inversions to prevent separation into two phases. If separation occurs, re-autoclave to re-homogenize.


- Store at 4°C.
